Biosynthesis of bacterial lipopolysaccharide. VII. Enzymatic formation of the first intermediate in biosynthesis of the O-antigen of Salmonella typhimurium.

چکیده
The initial step in biosynthesis of the O-antigen of Salmonella typhimurium is the formation of a lipid-linked derivative of galactose l-phosphate, according to the reaction UDP-galactose + P-lipid ti galactose-1 -PP-lipid + UMP. The enzyme activity has been detected in the cell envelope fraction of Citrobocter strain 139 (04, 5, 12) as well as in wild type S. typhimurium strain LT-2, but is most readily demonstrated in cell envelope fractions obtained from a mutant strain of S. fyphimurium lacking UDP-galactose 4epimerase following growth in the presence of galactose. The enzyme catalyzes transfer of the entire galactose lphosphate moiety of UDP-galactose to the lipid acceptor with concomitant formation of stoichiometric amounts of UMP. The reaction is readily reversible, with an apparent equilibrium constant of approximately 0.5.

Biosynthesis of a bacterial lipopolysaccharide. VI. Mechanism of incorporation of abequose into the O-antigen of Salmonella typhimurium.
Enzymatic incorporation of abequose into the repeating unit of the O-antigen is catalyzed by the particulate cell envelope fraction of Salmonella typhimurium. The initial step is the formation of a lipid-linked tetrasaccharide intermediate according to the reaction cytidine diphosphate abequose + mannosyl-rhamnosyl-galactose1 PP-lipid -+ abequosyl-mannosyl-rhamnosyl-galactose -1 -PP-lipid. The incomplete lipid A precursor produced by a mutant conditionally defective in synthesis of 3-deoxy-D-mannoctulosonate (KDO) is rapidly converted to lipopolysaccharide when the mutant culture is shifted from nonpermissive to permissive conditions (Osborn, M.J., Rick, P.D., and Rasmussen, N.S. (1980) J. Biol. Chem. 255, 4246-4251). An intermediate product which accumulates transiently during t...

S.typhimurium is a gram negative bacterium causing serious enteric or extraintestinal infections in human, cattle and birds. These kinds of infections may lead to some permanent or nonpermanent complications like endophthalmitis, Guillain-Barre syndrome and osteomyelitis . Multi-resistant strains of S.typhimurium have been reported from several countries, and these infections are a major public...
متن کاملAn intermediate step in translocation of lipopolysaccharide to the outer membrane of Salmonella typhimurium.
Evidence for transient localization of newly synthesized lipopolysaccharide at the periplasmic face of the inner membrane has been obtained by immunoelectron microscopic techniques. Salmonella typhimurium galE mutants in which O-antigen synthesis is dependent on addition of exogenous galactose were employed, and the distribution and fate of pulse-synthesized O antigen was examined by indirect f...
متن کاملAntibody Response to Bacterial Antigens: Characteristics of Antibody Response to Somatic Antigens of Salmonella typhimurium.
The character of the antibody response in the rabbit to Salmonella typhimurium somatic (O) antigen was similar to the response to each of several serotypes of Shigella flexneri O antigens, namely a predominance of production of immunoglobulin M (IgM) antibody. Lipopolysaccharide protein (LPSP) and lipopolysaccharide (LPS) fractions of Salmonella O antigen differed significantly in both quantita...
